Research Associate - Single molecule and super-resolution imaging of chromatin structure in mouse ES cells (Fixed Term)

Applications are invited for a Post-doctoral Research Associate who will be based in the Department of Chemistry, University of Cambridge to carry out single molecule super-resolution imaging of chromatin structure and its assembly. You will join an MRC- and Wellcome Trust-funded collaborative research project focusing on the relationship between chromatin structure and gene expression and understanding how the NuRD complex assembles and functions in mouse embryonic stem cells (mESC’s). The work is carried out in a close collaboration involving Prof Ernest Laue at the Department of Biochemistry, Dr Brian Hendrich at the Stem Cell Institute and Professor David Klenerman at the Department of Chemistry.

This exciting and multi-disciplinary project exploits super-resolution fluorescence microscopy and single nucleus Hi-C experiments to study nuclear architecture (see Stevens et al., Nature, 2017, 544, 59-64; Lando et al., Nature Protocols, 2018, 13, 1034-1061; Lando et al., Nucleus, 2018, 9, 190-201). The project uses tagged proteins with photo-activatable fluorophores, and studies these proteins in both fixed and live cells at the single molecule level. In particular, it will involve further development of single cell imaging methods to improve the resolution and rate of data acquisition as well as the application of the current method.

The post would suit candidates with a strong background in single cell imaging and/or super- resolution microscopy and a desire to perform cutting edge multi-disciplinary research. The successful candidate should have a PhD (or be close to completion). It is also highly desirable that they have experience in building or automating a single molecule or super-resolution microscope.

Fixed-term: The funds for this post are available for 1 year in the first instance, with the possibility of further funding for 2 more years.
